Procedeu de obţinere a bioetanolului din deşeuri agroalimentare tratate cu enzime termostabile recombinate, obţinute prin expresie heterologă extracelulară

Abstract: Abstract În procedeele industriale de prelucrare în alcool etilic a diferitelor produse agricole și/sau deșeuri de această proveninență, etapa cheie în definirea randametului procesului constă în eficiența hidrolizei diferitelor polizaharide în monozaharide fermentabile. În prezenta lucrare se propune obținerea unor enzime de înaltă eficiență pentru hidroliza amidonului caracteristic din cartofi, prin metodele ingineriei genetice. În prima etapă a lucrării s-a realizat un screening al diferitelor amilaze, ce se găsesc în microorganisme gazdă ce funcționează în medii extremofile. S-au selectat astfel trei tipuri de enzime: alfa amilaza din bacteria Bacillus licheniformis, o glucoamilază din fungia Aspergillus awamori, respectiv pullulanaza din Fervidobacter pennivorans, a căror gene au fost izolate din microorganismele gazdă, respectiv sintetizate în baza codului genetic cunoscut. În a doua parte a lucrării au fost sintetizate enzimele numite prin expresie heterologă, în diferite tulpini de microorganisme. În acest scop, prin metodele ingineriei genetice s-au construit vectorii purtători a genelor menționate, respectiv purificarea prin cromatografie a enzimelor obținute. În ultima parte a tezei s-au verificat proprietățile enzimelor și s-a propus o tehnologie privind utilizarea acestora într-un proces industrial. Abstract In the industrial processing of various agricultural products and/or different wastes of these origins to ethanol, the key stage in defining the yield of the process consists of the hydrolyzation efficiency of different polysaccharides to fermentable monosaccharides. In present work, the obtaining through genetic engineering of enzymes with higher efficiency for hydrolysis of the characteristical starch of potato is proposed. In the first stage of the work the screening of different amylases is realized, which are found in host microorganisms from extremophile environments. Three types of enzymes were selected this way: Bacillus licheniformis alpha-amylase, a glucoamylase from the Aspergillus awamori fungi, and the Fervidobacter pennivorans pullulanase, whose gene were isolated from their host microorganisms, respectively they were synthesized basing on their known genetic code. In the second part of the work the enzymes were synthesized with heterologous expression from different microorganism strains. For this purpose, with genetic engineering methods the carrier vectors of the mentioned genes were built, respectively the chromatographic purification of the obtained enzymes was realized. In the last part of the work the characterization of the enzymes was carried out and a technological suggestion was made on the utilization of these enzymes in an industrial process.
