OBTINEREA SI CARACTERIZAREA FIZICO-CHIMICA A UNOR EXTRACTE SELECTIVE DE NATURA VEGETALA CU APLICATII ÎN TERAPIA AFECTIUNILOR GASTRO-INTESTINALE

Abstract: OBTINEREA SI CARACTERIZAREA FIZICO-CHIMICA A UNOR EXTRACTE SELECTIVE DE NATURA VEGETALA CUAPLICATII ÎN TERAPIA AFECTIUNILOR GASTRO-INTESTINALE Obiectivele prezentei lucrari constau în obtinerea, caracterizarea fizico-chimica si testarea farmacotoxicologica preclinica a doua produse naturale de origine vegetala constituite din extracte selective îmbogatite în compusi vegetali cu potential gastro-protector si, respectiv, anticolitic ridicat. Produsul farmacologic activ cu potential gastroprotector se prezinta ca o pulbere bruna total solubila în dimetilsulfoxid compusa din: flavonozide cu potential gastroprotector ridicat (derivati ai quercetinului, luteolinului si apigeninei nesubstituiti la nucleul fenil); acizi polifenolcarboxilici cu potential antioxidant puternic, reglatori ai secretiei biliare (acid cafeic, acid clorogenic si acid izoclorogenic); polizaharide cu bioadezivitate mare la mucoasa gastrica, minerale si proteine. Produsul gastroprotector este standardizat în compusii vegetali cu activitate farmacologica cheie: 3,2% flavone totale exprimate în rutin si 3,7% polifenoli totali exprimati în acid cafeic (g). Produsul farmacologic activ cu potential anticolitic se prezinta ca o pulbere de culoare galben-bruna total solubila în dimetilsulfoxid care contine: quercetin si derivati ai acestuia glicozilati la fenil (quercetin-3'/4'-glicozide) cu potential spasmolitic-antioxidant-antiinflamator marcat la nivel intestinal, plus polizaharide specifice (pectine) si minerale capabile sa reduca chemochineza si concentratia de toxine de la acest nivel. Produsul anticolitic este standardizat în flavone totale exprimate în rutin: 10g%. Studiile farmacotoxicologice preclinice in vivo au demonstrat faptul ca produsele obtinute pot fi încadrate în categoria “practic netoxice”, iar studiile privind activitatea specifica au confirmat potentialul terapeutic ridicat al acestora. Ca urmare, prezenta lucrare constituie un model de obtinere a unor produse medicamentoase naturale constituite din extracte selective de natura vegetala pornind de la particularitatile biochimice ale maladiei, cumulat cu corelatia dintre structura compusilor naturali si functia lor biologica. OBTAINING AND CHEMICAL CHARACTERIZATION OF SOME SELECTIVE PLANT EXTRACTS WITH APPLICATIONS IN THE THERAPY OF GASTRO-INTESTINAL DISEASES This work aims the obtaining and physical-chemical and non-clinical pharmacological characterization of two plant extracts composed of selective extracts enriched in vegetal compounds with high gastro-protective and, respectively, intestinal healing properties. Gastro-protective plant extract is presented as a brown powder soluble into dimethilsulfoxide. It contains: flavones glycosides with known gastro-protective activity (quercetin, luteolin and apigenin glycosides with free OH groups at phenil ring); phenolcarboxilic acids with high antioxidant properties able to restore gall bladder activity (caffeic, chlorogenic and, respectively, izochlorogenic acids); polysaccharides with strong bio-adhesive properties on mucous gastric tissue, minerals and proteins. Gastro-protective product is standardized as key compounds: 3.2% total flavones expressed as rutin and, respectively, 3.7% total phenols expressed as caffeic acid (g). Vegetal product with healing properties on intestinal damaged tissue is presented as a yellow-brown powder soluble into dimethilsulfoxide. It contains quercetin and quercetin glycosides at phenil ring (3'/4'-quercetin glycosides) with strong non-spastic, antioxidant, anti-inflammatory properties at the intestinal level and, also, specific polysaccharides (pectins) and minerals capable to reduce chemochinesis and toxin level. Vegetal product is standardized as flavones expressed in rutin: 10g%. Pharmacological non-clinical in vivo studies revealed these two vegetal products as being non-toxic, having strong gastro-protective and, respectively, intestinal healing properties, as well. Consequently, this work can be used as a model of obtaining new, natural drugs based on selective plant extracts taking into account of biochemical particularities of diseases, correlated with structure/function relationship.
